The Vedek Assembly was the governing body of the Bajoran religion and held great political influence as well. The Assembly was led by the kai, the leading religious figure on Bajor.

In 2369, Commander Benjamin Sisko asked Vedek Bareil Antos to grant him an audience with the Assembly to discuss the school problem aboard Deep Space 9. Bareil refused and told Sisko that the Assembly feared him because of his role as Emissary of the Prophets, his position as Starfleet commander, and because Vedek Winn Adami told them to. In 2370, Jaro Essa intended to direct the Vedek Assembly to name Vedek Winn as kai twenty-six hours after he was sworn into office as part of his coup d'etat against the Bajoran Provisional Government. (DS9: "The Siege")

The Chamber of Ministers debated the request of the Skrreeans that they be allowed immigration to Bajor. Acting in accordance with the Vedek Assembly, the ministers denied the request. (DS9: "Sanctuary")

Prylar Rhit's sizable gambling debts from playing dabo had caused a scandal in the Vedek Assembly. (DS9: "Shadowplay")

Vedek Winn suggested that she and Sisko could address the Assembly together. She made the suggestion, according to her, as a celebration of their friendship. Sisko agreed, saying it was an excellent proposal, and that he could do it sometime next week. If he did it sooner, before the Bajoran Choosing Ceremony, he thought it might be misconstrued as an endorsement of Winn's bid for kai. As a Federation officer, he could not interfere in the internal affairs of Bajor. They came to an agreement that they would appear together before the Assembly sometime the following week. In 2371, the Vedek Assembly endorsed the Bajoran-Cardassian Treaty, and defrocked vedeks who publicly disagreed with that position. Vedek Yarka was one person so defrocked. (DS9: "Destiny")

In 2374, Kai Winn was aghast that Captain Sisko had removed an ancient tablet from the ruins of B'hala without consulting the Vedek Assembly. (DS9: "The Reckoning")

In late 2375, Winn Adami canceled an appearance before the Assembly. What was unknown at the time was that she had turned away from the Prophets and had embraced the Pah-wraiths. (DS9: "The Changing Face of Evil")
